CD301b +DCs orchestrate type 2 immune responses to helminth infection in the lung

Abstract Helminth parasites induce potent host type 2 immune responses (T2I) that promote parasite clearance and prevent reinfection. The parasite-induced T2I involve both innate adaptive cell components, but how the activation of different types is coordinated during initiation incompletely understood. Dendritic cells (DCs) are best known for their role in bridging immunity by sensing immunological insults presenting antigens to T cells. We previously found CD301b +DCs, a major subset migratory conventional DCs peripheral organs, specifically required differentiation helper (Th2) skin- lung-draining lymph nodes following infection with Nippostrongylus brasiliensis (Nb) mice, overall against Nb remains unclear. Here, we show depletion +DCs results reduced lung impaired worm clearance. reduction cytokine production eosinophil influx was observed as early days post-infection before onset immunity, well CD4T cell-depleted suggesting play critical induction independently Th2 priming. Moreover, +DC-depleted mice exhibited ameliorated hemorrhage neutrophil infiltration IL-17 after infection. These data suggest co-regulating 3 inflammation mechanisms phase lung.
